In-Cell Solid-State NMR Analysis of Membrane Proteins

Abstract
Solid-state NMR spectroscopy (ssNMR) has great potential to provide atomistic detail of membrane proteins (MPs) as it is suitable for highly heterogeneous and complex molecular environments, without the need to solubilize or crystallize the sample. ssNMR is able to determine inter and intramolecular distances, probe site-specific molecular dynamics, and obtain orientational structural restraints for membrane proteins in their native membranes or in near-native conditions using model membranes. Recent developments in dynamic nuclear polarization (DNP) techniques and ssNMR spectroscopy have led to the feasibility of in-cell NMR of MPs.
